Overview

A combination investigational anticancer agent in which **titanocene** (usually as titanocene dichloride), an organometallic titanium(IV) complex, is delivered together with **transferrin**, a natural iron-transport glycoprotein, and **titanium dioxide (TiO₂)**, an inert oxide of titanium. The rationale is that transferrin may facilitate the targeted delivery of titanium ions, derived from titanocene or TiO₂, to tumor cells that overexpress transferrin receptors, potentially increasing selective cytotoxicity. After systemic administration, titanocene rapidly interacts with transferrin, releasing its ligands and binding at transferrin’s iron (Fe(III)) sites, resulting in a Ti(IV)-transferrin complex. Titanium-loaded transferrin is taken up by tumor cells via transferrin receptor-mediated endocytosis. Titanium compounds (from titanocene or TiO₂) are experimental anticancer agents, distinct from platinum-based chemotherapies, and are under investigation for their activity against cisplatin-resistant tumors, though instability and rapid hydrolysis in vivo limit their efficacy. The therapeutic mechanism is believed to involve disruption of cellular redox balance, induction of apoptosis, and interference with DNA replication and repair, though it is not fully elucidated[1][2][5][6].
