Overview

Tivozanib + durvalumab is a combination therapy being evaluated for the treatment of advanced or metastatic hepatocellular carcinoma (HCC). Tivozanib is an oral, once-daily, selective tyrosine kinase inhibitor (TKI) of vascular endothelial growth factor receptors (VEGFR) 1, 2, and 3, designed to inhibit angiogenesis in tumors. Durvalumab is a fully human monoclonal antibody targeting programmed death-ligand 1 (PD-L1), acting as an immune checkpoint inhibitor to enhance antitumor immune responses. The combination therapy aims to inhibit tumor angiogenesis via VEGFR blockade (tivozanib) and augment immune-mediated tumor destruction (durvalumab). The combination has shown tolerable safety and promising preliminary efficacy in phase 1b/2 clinical trials for patients with advanced HCC, particularly those who are treatment-naïve, and is being further investigated in clinical trials[1][2][3][4][5].
