Overview

Tivozanib + paclitaxel is a combination of two anticancer agents used in clinical research for the treatment of solid tumors, including metastatic breast cancer. Tivozanib is an oral, potent, and selective small-molecule tyrosine kinase inhibitor that targets vascular endothelial growth factor receptors (VEGFR) 1, 2, and 3. By inhibiting these receptors at subnanomolar concentrations, tivozanib blocks angiogenesis and lymphangiogenesis critical for tumor growth[6][8]. Paclitaxel is a taxane chemotherapeutic agent that binds to the β subunit of tubulin in microtubules, hyper-stabilizing their structure and preventing normal cell division[5]. The combination has been studied in phase I trials showing tolerability and preliminary antitumor activity in metastatic breast cancer patients who had not previously received VEGFR inhibitors[3][8].
