Tivozanib + temsirolimus is a combination therapy investigated for the treatment of advanced renal cell carcinoma (RCC). Tivozanib is a potent and selective small molecule tyrosine kinase inhibitor targeting vascular endothelial growth factor receptors (VEGFR)-1, -2, and -3, thereby inhibiting tumor angiogenesis and growth. Temsirolimus is an mTOR inhibitor that binds to FKBP12 to inhibit the mammalian target of rapamycin pathway, resulting in cell cycle arrest at G1 phase and reduced synthesis of proteins involved in proliferation, survival, and angiogenesis. The combination has demonstrated acceptable tolerability with manageable toxicity in early-phase clinical studies for RCC[2][3][5].
