Drug intelligence / Profile preview
**tivunatamig + carboplatin + nab-paclitaxel** is an investigational combination regimen consisting of tivunatamig (PF-08634404 / SSGJ-707), a bispecific tetravalent monoclonal antibody targeting programmed cell death protein 1 (PD-1) and vascular endothelial growth factor A (VEGF-A), combined with the platinum-based chemotherapeutic carboplatin and the nanoparticle albumin-bound formulation of paclitaxel (nab-paclitaxel). Tivunatamig was originally developed by Dansheng Pharmaceutical Technology, a subsidiary of Sunshine Guojian and 3SBio, and was licensed globally (excluding China) to Pfizer in 2025. The combination is designed to leverage the synergistic effects of immune checkpoint blockade, anti-angiogenesis, and direct cytotoxicity. By blocking PD-1, tivunatamig reactivates T-cell-mediated anti-tumor immunity, while its anti-VEGF-A activity inhibits tumor angiogenesis and reverses VEGF-mediated immunosuppression in the tumor microenvironment. Carboplatin induces DNA crosslinking to trigger apoptosis, and nab-paclitaxel disrupts microtubule dynamics to inhibit cell division. This triplet regimen is primarily being evaluated in Phase II and Phase III clinical trials as a first-line treatment for advanced or metastatic squamous non-small cell lung cancer (NSCLC).
