Overview

TLK286 (also known as Telcyta) is an investigational antitumor prodrug developed by Telik. It is designed to be selectively activated in tumor cells that overexpress glutathione S-transferase P1-1 (GSTP1), an enzyme associated with drug resistance and poor prognosis in various solid tumors, including ovarian, lung, breast, gastrointestinal, and head and neck cancers. Upon activation by GSTP1 within the tumor cell, TLK286 initiates apoptosis specifically in these cancer cells[1]. Docetaxel is a well-established taxane-class chemotherapeutic agent that works by promoting microtubule assembly and stabilizing them against depolymerization, thereby inhibiting cell division and inducing apoptosis in rapidly dividing cancer cells[2][4]. The combination of TLK286 with docetaxel has been investigated clinically for enhanced efficacy against cancers such as non-small cell lung cancer (NSCLC) and ovarian cancer[1].
