**TmAb-MLINK Duo-MMAE + exatecan** is a preclinical, trastuzumab-based dual-payload antibody-drug conjugate developed by Debiopharm using its MLINK Duo linker technology. The HER2-directed trastuzumab antibody delivers monomethyl auristatin E, a microtubule-disrupting payload, and exatecan, a topoisomerase I-inhibitor payload. The MLINK Duo platform is designed to provide high plasma stability and tumor-specific enzymatic release of both payloads; in cell-derived xenograft models, this construct showed antitumor activity and tumor regression in models resistant to corresponding single-payload MMAE or exatecan ADCs.
