TMV-018 + 5-FC is an investigational oncolytic virotherapy combination. TMV-018 is a recombinant measles virus (Schwarz strain) engineered to express a 'super cytosine deaminase' (SCD) transgene. When administered intratumorally, the virus selectively replicates in and lyses cancer cells. The SCD enzyme expressed by the virus converts the co-administered prodrug 5-fluorocytosine (5-FC) into the potent chemotherapeutic agent 5-fluorouracil (5-FU) locally within the tumor microenvironment. This dual approach combines direct viral oncolysis with targeted local chemotherapy to enhance anti-tumor activity while minimizing systemic toxicity. It is being developed by Themis Bioscience (a subsidiary of Merck & Co.) for the treatment of advanced gastrointestinal malignancies, including colorectal, esophageal, and gastric cancers.
