**TNFerade + cetuximab** is an investigational combination regimen composed of TNFerade, a gene therapy product encoding human tumor necrosis factor-alpha (TNF-α), and cetuximab, a recombinant chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR). TNFerade delivers a therapeutic transgene to tumor sites by adenoviral vector, leading to local production of TNF-α, which acts as a potent pro-inflammatory and pro-apoptotic cytokine, promoting antitumor effects and sensitization to other therapies. Cetuximab binds EGFR and blocks ligand-induced activation, inhibiting downstream signaling that promotes tumor cell proliferation and survival. The combination has been investigated in cancer clinical trials under the hypothesis that TNFerade’s pro-inflammatory environment may boost the cytotoxicity or immune effects of cetuximab and potentially enhance overall antitumor activity. These agents are not approved as a fixed combination and have not received regulatory approval as a combination product; the combination is considered investigational.
