TNG260 + pembrolizumab

This is an investigational combination therapy consisting of **TNG260**, a small-molecule, orally available inhibitor of the CoREST (Co-repressor of Repressor Element-1 Silencing Transcription) complex—including selective inhibition of HDAC1—and **pembrolizumab**, a humanized monoclonal antibody that targets PD-1. TNG260 is designed to reverse resistance to immune checkpoint inhibitor therapy in cancers with STK11 mutations by remodeling the tumor microenvironment, reducing immunosuppressive neutrophil presence, increasing T cell recruitment, and restoring sensitivity to anti-PD-1 blockade. Pembrolizumab blocks PD-1/PD-L1/PD-L2 interaction, augmenting the antitumor T cell response. The combination is currently under clinical evaluation in STK11-mutated, locally advanced or metastatic solid tumors, for example non-small cell lung cancer, breast, pancreatic, cervical, endometrial, and carcinomas of unknown primary[1][2][3][4][5][6][7].
