Overview

TNO155 + ribociclib is a combination of two small molecule inhibitors evaluated for treatment of advanced solid tumors. **TNO155** is an oral, selective inhibitor of *SHP2* (Src homology region 2 domain-containing phosphatase-2), an intracellular phosphatase involved in the RAS/MAPK signaling pathway. Inhibition of SHP2 can suppress tumor growth, particularly in cancers with aberrant MAPK pathway activation. **Ribociclib** is an oral cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitor, approved for hormone receptor-positive, HER2-negative advanced breast cancer, that blocks cell cycle progression by preventing phosphorylation of the retinoblastoma protein. Preclinical models and early clinical trials suggest the combination may exhibit enhanced anti-tumor activity compared to either agent alone, likely due to combined blockade of oncogenic signaling and cell cycle progression. Novartis is the principal developer of both agents, and the combination was studied in phase 1b trials in advanced solid tumors[1][2][3].
