Overview

TNO155 + spartalizumab is an investigational combination therapy for advanced solid tumors. TNO155 is a small molecule inhibitor of SHP2 (Src homology region 2-containing protein tyrosine phosphatase), which plays a key role in RTK- and MAPK-driven oncogenic signaling, impacting tumor growth and immune evasion. Spartalizumab (PDR001) is a monoclonal antibody targeting programmed cell death protein 1 (PD-1), acting as an immune checkpoint inhibitor to relieve suppression of anti-tumor immune responses. This combination is being developed to inhibit both tumor-promoting pathways and immunosuppressive signaling, aiming to improve clinical outcomes over single-agent therapies[1][3][5].
