Overview

TNP-2092 + vancomycin is a combination of two antibacterial agents: TNP-2092, an investigational hybrid antibiotic that fuses rifamycin and quinolizinone pharmacophores, and vancomycin, a well-established glycopeptide antibiotic. This combination has been studied for the treatment of acute and device-associated skin and soft tissue infections, as well as prosthetic joint infections, particularly those caused by Gram-positive bacteria. TNP-2092 acts via inhibition of DNA gyrase, DNA topoisomerase, and DNA-directed RNA polymerase, thereby disrupting bacterial DNA replication and transcription[2][3][5][7]. Vancomycin inhibits bacterial cell wall synthesis by binding to the D-Ala-D-Ala terminus of cell wall peptide precursors. The combination is primarily developed by TenNor Therapeutics, with TNP-2092 designated as a new molecular entity and awarded orphan drug status for bacterial infections[2].
