Overview

A combination of two biologic disease-modifying antirheumatic drugs (DMARDs) with different mechanisms of action. Tocilizumab is an interleukin-6 (IL-6) receptor antagonist that blocks IL-6 signaling by binding to soluble and membrane-bound IL-6 receptors. Abatacept is a fusion protein that selectively modulates T-cell activation by binding to CD80/CD86, blocking interaction with CD28. This inhibits the costimulatory signal required for full T-cell activation, which is implicated in the autoimmune response seen in rheumatoid arthritis. This combination is not typically administered simultaneously due to increased risk of adverse effects, though each drug individually is effective for rheumatoid arthritis treatment.
