Overview

A combination of tolfenamic acid, a non-steroidal anti-inflammatory drug (NSAID) with anticancer properties, and gemcitabine, a nucleoside metabolic inhibitor. Tolfenamic acid works through COX-independent mechanisms by degrading specificity protein transcription factors (Sp1, Sp3, and Sp4) and reducing VEGF expression, which inhibits tumor growth and angiogenesis. Gemcitabine is a cytidine analog that works by replacing building blocks of nucleic acids during DNA elongation, arresting tumor growth and promoting apoptosis of malignant cells. This combination is being investigated for the treatment of pancreatic cancer, particularly in combination with radiation therapy.
