Tomuzotuximab + chemotherapy is a combination therapeutic regimen consisting of **tomuzotuximab**, a glyco-engineered monoclonal antibody targeting the epidermal growth factor receptor (EGFR), administered together with standard chemotherapy agents. Tomuzotuximab is engineered to enhance antibody-dependent cellular cytotoxicity (ADCC) by modifying its glycosylation profile, resulting in increased efficacy against EGFR-expressing tumor cells. In clinical studies, it is commonly combined with platinum-based chemotherapy (cisplatin) and fluorouracil (5-FU) for the treatment of recurrent or metastatic squamous cell carcinoma of the head and neck. The rationale for the combination is that tomuzotuximab blocks EGFR signaling to inhibit tumor growth and survival, while chemotherapy induces cytotoxic damage to rapidly dividing cancer cells[4][9]. This combination has been evaluated in randomized phase II trials but did not show superiority over cetuximab plus chemotherapy as first-line treatment for this indication[4].
