Overview
Toripalimab + axitinib is a combination therapy consisting of two agents: toripalimab, a humanized IgG4 monoclonal antibody that targets programmed cell death protein 1 (PD-1), and axitinib, a small molecule tyrosine kinase inhibitor targeting vascular endothelial growth factor receptors (VEGFRs). Toripalimab acts as an immune checkpoint inhibitor by blocking PD-1, thereby enhancing T-cell mediated antitumor immunity. Axitinib inhibits angiogenesis and tumor growth by selectively blocking VEGFRs. This combination has demonstrated synergistic antitumor activity in clinical trials for advanced renal cell carcinoma (RCC) and metastatic mucosal melanoma. In China, it is approved for first-line treatment of intermediate to high-risk unresectable or metastatic RCC. The regimen has also shown promising efficacy in neoadjuvant settings for clear cell RCC and in patients with advanced mucosal melanoma[2][3][4][7][8][9].
