JS001 + carboplatin + cisplatin + etoposide is a combination chemo-immunotherapy regimen developed by Shanghai Junshi Biosciences for the first-line treatment of extensive-stage small cell lung cancer (ES-SCLC). The regimen consists of toripalimab (JS001), a humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, combined with etoposide and a platinum agent (either carboplatin or cisplatin). By blocking the PD-1/PD-L1 pathway, toripalimab restores T-cell anti-tumor activity, while the chemotherapy components provide direct cytotoxic effects and potentially enhance immunogenicity. This combination was approved by China's NMPA in June 2024 based on the Phase 3 EXTENTORCH trial, which demonstrated significant improvements in progression-free survival and overall survival compared to chemotherapy alone.
