This combination therapy is a neoadjuvant regimen consisting of the PD-1 inhibitor toripalimab, the platinum-based chemotherapy carboplatin, and the albumin-bound microtubule inhibitor nab-paclitaxel. It is currently being investigated in Phase II clinical trials by Sun Yat-Sen Memorial Hospital for the treatment of resectable salivary gland malignant neoplasms. Toripalimab, developed by Shanghai Junshi Bioscience, is a humanized IgG4 monoclonal antibody that blocks the interaction between PD-1 and its ligands (PD-L1 and PD-L2), thereby restoring T-cell mediated anti-tumor immunity. Carboplatin induces cell death by creating interstrand DNA cross-links, while nab-paclitaxel inhibits mitosis by stabilizing microtubules. The combination aims to reduce tumor size and improve surgical outcomes through synergistic immune activation and cytotoxic effects.
