Toripalimab + cetuximab is an investigational combination therapy consisting of two monoclonal antibodies: toripalimab, a humanized IgG4 monoclonal antibody targeting programmed cell death protein 1 (PD-1), and cetuximab, a chimeric IgG1 monoclonal antibody targeting the epidermal growth factor receptor (EGFR). Toripalimab blocks the PD-1/PD-L1 pathway, thereby enhancing T-cell-mediated immune responses against tumor cells. Cetuximab inhibits EGFR signaling, which is often upregulated in various cancers and promotes tumor cell proliferation and survival; it also induces antibody-dependent cellular cytotoxicity. The combination aims to provide synergistic antitumor effects by simultaneously blocking immune checkpoint inhibition and EGFR-driven oncogenic signaling. This regimen has shown promising efficacy and manageable safety in clinical trials for recurrent or metastatic head and neck squamous cell carcinoma (R/M HNSCC), particularly in patients with PD-L1-positive tumors[1][3][5].
