Overview
Toripalimab + cetuximab + platinum is a combination regimen under clinical investigation for the treatment of recurrent or metastatic head and neck squamous cell carcinoma (R/M HNSCC), particularly in patients who are refractory to or have progressed after platinum-based chemotherapy. Toripalimab is a humanized IgG4 monoclonal antibody that targets programmed cell death protein 1 (PD-1), acting as an immune checkpoint inhibitor to enhance anti-tumor immune responses. Cetuximab is an IgG1 monoclonal antibody targeting the epidermal growth factor receptor (EGFR), inhibiting tumor cell proliferation and survival, and also mediating antibody-dependent cellular cytotoxicity. Platinum agents (such as cisplatin or carboplatin) are DNA-damaging chemotherapeutic drugs that induce apoptosis in rapidly dividing cells. The rationale for this combination lies in the potential synergy between immunotherapy, targeted therapy, and cytotoxic chemotherapy to improve outcomes in R/M HNSCC[1][2][7].
