A combination therapy consisting of toripalimab, an anti-PD-1 monoclonal antibody, and chidamide (tucidinostat), a selective histone deacetylase (HDAC) inhibitor. This combination works through dual mechanisms: toripalimab blocks the PD-1/PD-L1 pathway to activate the body's immune system against cancer cells, while chidamide modulates abnormal epigenetic regulation by inhibiting specific HDAC subtypes (1, 2, 3, and 10), leading to increased histone acetylation, cell cycle arrest, and apoptosis in tumor cells. Chidamide has also been shown to increase PD-L1 expression and enhance CD8+ T cell infiltration in the tumor microenvironment, providing a synergistic effect with anti-PD-1 therapy.
