This combination therapy consists of toripalimab, a humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, and dasatinib, a small-molecule multi-tyrosine kinase inhibitor. The regimen is currently being investigated in a Phase 2 clinical trial led by The Affiliated Hospital of Qingdao University as a third-line treatment for patients with advanced or stage IV non-small cell lung cancer (NSCLC) harboring ARID1A mutations. Toripalimab functions as a checkpoint inhibitor, blocking the interaction between PD-1 and its ligands (PD-L1 and PD-L2) to restore T-cell mediated anti-tumor immunity. Dasatinib, which targets BCR-ABL, SRC family kinases, c-KIT, and other tyrosine kinases, is hypothesized to synergize with PD-1 blockade, particularly in tumors with ARID1A deficiency which may exhibit specific vulnerabilities to kinase inhibition or altered immune microenvironments.
