toripalimab + fruquintinib + short-course radiotherapy is a neoadjuvant combination therapy regimen being investigated for the treatment of locally advanced rectal cancer (LARC). The regimen consists of toripalimab, a humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor; fruquintinib, a highly selective oral tyrosine kinase inhibitor of vascular endothelial growth factor receptors (VEGFR) 1, 2, and 3; and short-course radiotherapy (SCRT). This combination aims to enhance anti-tumor activity through the synergistic effects of immune checkpoint blockade, inhibition of tumor angiogenesis, and radiation-induced immunogenic cell death. The regimen is currently being evaluated in a Phase II clinical trial led by West China Hospital of Sichuan University to assess its efficacy and safety in achieving pathological complete response in patients with LARC.
