Overview

The PraG strategy is an investigational combination immunotherapy regimen being evaluated for the treatment of soft tissue sarcoma. It consists of the programmed cell death protein 1 (PD-1) inhibitor toripalimab, recombinant human granulocyte-macrophage colony-stimulating factor (GM-CSF), and the immunomodulator thymalfasin, administered in conjunction with spatially fractionated radiotherapy (SFRT). Developed and studied by the First Affiliated Hospital of Guangxi Medical University, this multi-modal approach aims to enhance the systemic anti-tumor immune response (the abscopal effect) by combining checkpoint inhibition with cytokine-driven dendritic cell maturation and radiation-induced antigen release. Toripalimab, developed by Junshi Biosciences, serves as the backbone of the regimen by preventing PD-1 mediated immune evasion, while GM-CSF and thymalfasin act as immunomodulators to stimulate the production and activation of immune cells.
