Toripalimab + interferon alpha is an investigational combination immunotherapy being evaluated for the treatment of head and neck squamous cell carcinoma (HNSCC). Toripalimab is a humanized monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, blocking its interaction with ligands PD-L1 and PD-L2 to restore T-cell mediated anti-tumor activity. Interferon alpha is a cytokine that enhances the immune response by promoting the maturation of dendritic cells, increasing MHC class I expression, and potentially overcoming resistance to PD-1 blockade. This combination is being studied, notably by the Ninth People's Hospital Affiliated to Shanghai Jiao Tong University, to improve response rates in patients with recurrent or metastatic HNSCC who may have limited options with standard therapies.
