Overview

Toripalimab-Sun Yat-sen University-hepatocellular carcinoma refers to a specific combination therapy regimen investigated by Sun Yat-sen University Cancer Center for the treatment of advanced hepatocellular carcinoma (HCC). The regimen integrates systemic immunotherapy with toripalimab (a humanized IgG4 monoclonal antibody targeting PD-1), systemic targeted therapy with lenvatinib (a multi-kinase inhibitor), and localized hepatic arterial infusion chemotherapy (HAIC) using the FOLFOX protocol (oxaliplatin, leucovorin, and 5-fluorouracil). This approach is designed to overcome the limitations of monotherapy in advanced HCC, particularly in patients with high-risk features like portal vein tumor thrombosis (PVTT). Clinical trials, including Phase 2 and Phase 3 studies, have demonstrated that this triple combination significantly improves progression-free survival (PFS) and overall survival (OS) compared to standard-of-care treatments by combining immune activation, anti-angiogenesis, and direct cytotoxic tumor debulking.
