Overview
Toripalimab (JS001) is a humanized IgG4 monoclonal antibody developed by Shanghai Junshi Biosciences that targets the programmed cell death protein 1 (PD-1) receptor. As an immune checkpoint inhibitor, it blocks the interaction between PD-1 and its ligands (PD-L1 and PD-L2), thereby restoring the anti-tumor activity of T cells. In a specific Phase II clinical trial sponsored by Wan He, toripalimab is being evaluated as a perioperative treatment in combination with the FLOAP chemotherapy regimen. The FLOAP regimen consists of nanoparticle albumin-bound (nab) paclitaxel, oxaliplatin, fluorouracil (5-FU), and leucovorin. This combination therapy is being investigated for patients with locally advanced gastric cancer and gastroesophageal junction (GEJ) adenocarcinoma, with the primary objective of increasing the pathological complete response (pCR) rate and improving long-term survival outcomes.
