Toripalimab + JS002 is an investigational combination therapy developed by Shanghai Junshi Bioscience Co., Ltd. It consists of toripalimab (JS001), a recombinant humanized monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, and ongericimab (JS002), a recombinant humanized monoclonal antibody targeting proprotein convertase subtilisin/kexin type 9 (PCSK9). While PCSK9 inhibitors are primarily used for lipid-lowering indications, this combination was explored in oncology based on research suggesting that PCSK9 inhibition can increase the cell surface expression of MHC I, potentially synergizing with PD-1 blockade to enhance anti-tumor T-cell responses. A Phase I clinical trial (NCT05128539) was conducted to evaluate the safety and preliminary efficacy of the combination in patients with advanced cancer; however, the study was terminated early because the efficacy did not reach the predefined threshold.
