toripalimab + paclitaxel + cisplatin

Toripalimab + TP is a combination therapy consisting of toripalimab, a humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, and the chemotherapy doublet TP, which comprises paclitaxel and cisplatin. Toripalimab functions as an immune checkpoint inhibitor by blocking the interaction between PD-1 and its ligands (PD-L1 and PD-L2), thereby restoring T-cell mediated anti-tumor activity. Paclitaxel is a taxane that stabilizes microtubules, leading to mitotic arrest and apoptosis, while cisplatin is a platinum-based agent that forms DNA cross-links, interfering with DNA replication and transcription. This combination is primarily investigated as a neoadjuvant treatment for locally advanced head and neck squamous cell carcinoma (HNSCC), aiming to reduce tumor burden and improve surgical outcomes through synergistic cytotoxic and immunomodulatory effects.
