Overview
Toripalimab + pazopanib is a combination therapy being evaluated for the treatment of advanced clear cell renal cell carcinoma (ccRCC). Toripalimab is a humanized IgG4 monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, blocking its interaction with PD-L1 and PD-L2 to enhance anti-tumor T-cell responses. Pazopanib is an orally active, multi-target tyrosine kinase inhibitor (TKI) that inhibits vascular endothelial growth factor receptors (VEGFR-1, -2, and -3), platelet-derived growth factor receptors (PDGFR-α and -β), and the stem cell factor receptor (c-kit), thereby suppressing tumor angiogenesis and cell proliferation. This combination regimen, notably investigated by the Fudan University Shanghai Cancer Center, aims to provide a synergistic therapeutic effect by combining immune checkpoint inhibition with anti-angiogenic targeted therapy in patients with metastatic or advanced renal malignancies.
