Toripalimab + platinum-based doublet chemotherapy is a combination regimen comprising the anti-PD-1 monoclonal antibody toripalimab and a standard-of-care platinum-based chemotherapy doublet. Toripalimab, developed by Junshi Biosciences, is a humanized IgG4 monoclonal antibody that binds to the programmed cell death protein 1 (PD-1) receptor on T cells, blocking its interaction with ligands PD-L1 and PD-L2 to restore anti-tumor immune activity. The platinum-based doublet typically consists of a platinum agent (cisplatin or carboplatin) paired with a third-generation cytotoxic agent (such as pemetrexed, paclitaxel, or gemcitabine), which induces DNA damage and inhibits cell division. This combination has demonstrated significant efficacy in the perioperative setting for resectable Stage II-III non-small cell lung cancer (NSCLC), as shown in the Phase III NEOTORCH trial, and is also used in the first-line treatment of advanced NSCLC and other solid tumors.
