This combination therapy consists of toripalimab, a humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor; tiflamimab (JS004), a humanized monoclonal antibody targeting the B- and T-lymphocyte attenuator (BTLA); and docetaxel, a cytotoxic chemotherapy agent of the taxane class. Toripalimab works by blocking the interaction between PD-1 and its ligands (PD-L1 and PD-L2), thereby restoring T-cell mediated anti-tumor immune responses. Tiflamimab is a first-in-class anti-BTLA antibody that blocks the BTLA-HVEM (Herpesvirus Entry Mediator) interaction, which is an immune checkpoint pathway that suppresses T-cell activation. Docetaxel inhibits microtubule depolymerization, leading to cell cycle arrest and apoptosis. The combination is being evaluated, particularly by researchers at West China Hospital of Sichuan University, for the treatment of advanced non-squamous non-small cell lung cancer (NSCLC) in patients who have progressed on prior anti-PD-1/L1 therapies.
