Toripalimab + topotecan is a combination therapy consisting of toripalimab, a humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, and topotecan, a semi-synthetic derivative of camptothecin that acts as a topoisomerase I inhibitor. This combination is being evaluated in a Phase II clinical trial sponsored by Fudan University for the treatment of patients with advanced or recurrent endometrial cancer who have progressed after first-line therapy. Toripalimab works by blocking the interaction between PD-1 and its ligands (PD-L1 and PD-L2), thereby restoring T-cell mediated anti-tumor immune responses. Topotecan inhibits DNA replication by stabilizing the topoisomerase I-DNA complex, leading to double-strand DNA breaks and cell death. The combination aims to leverage the potential synergistic effects of chemotherapy-induced immunogenic cell death and checkpoint inhibition.
