Overview

Toripalimab + vorolanib is an investigational combination therapy consisting of two agents with complementary mechanisms of action for cancer treatment. Toripalimab is a humanized IgG4 monoclonal antibody that targets programmed death-1 (PD-1), functioning as an immune checkpoint inhibitor to enhance T-cell mediated anti-tumor responses. Vorolanib (CM082) is a small molecule multi-target tyrosine kinase inhibitor that blocks vascular endothelial growth factor receptors (VEGFRs), platelet-derived growth factor receptor (PDGFR), c-kit, and Flt-3, thereby inhibiting angiogenesis and tumor proliferation. The rationale for combining these drugs lies in the ability of VEGF inhibition to reduce immunosuppression in the tumor microenvironment and potentially overcome resistance to immune checkpoint blockade. This combination has been studied in phase II clinical trials for non-small cell lung cancer (NSCLC) and advanced mucosal melanoma, showing promising antitumor activity with acceptable safety profiles[1][2][3][4].
