tosatoxumab + standard of care antibiotics

Tosatoxumab + standard of care antibiotics is a combination therapy consisting of **tosatoxumab** (AR-301), a fully human monoclonal IgG1 antibody that specifically targets and neutralizes Staphylococcus aureus alpha-toxin (hemolysin A, Hla), together with standard of care antibiotics. This regimen is investigated as an adjunctive treatment for ventilator-associated pneumonia (VAP) caused by S. aureus (including MRSA and MSSA). Tosatoxumab binds to an N-terminal epitope of alpha-toxin, preventing its pore-forming actions and cytotoxic effects, thereby protecting host cells and enhancing bacterial clearance. The antibody’s mechanism is independent of the antibiotic resistance profile of S. aureus, offering a novel passive immunotherapy approach.
