**Tosedostat + paclitaxel** is a combination regimen of two antineoplastic drugs used in investigational therapy for advanced solid tumors. **Tosedostat** (CHR-2797) is an orally administered, bioavailable aminopeptidase inhibitor that interferes with intracellular protein metabolism, inducing antiproliferative and proapoptotic effects, and displays antiangiogenic activity. **Paclitaxel** is a well-established intravenous chemotherapeutic agent that stabilizes microtubules, preventing cell division, and leading to apoptosis. In clinical studies, this combination showed tolerable toxicity but was associated with a high incidence of paclitaxel-related infusion reactions, possibly influenced by tosedostat. Antitumor activity was observed in a subset of patients, with instances of partial responses and stable disease over several months.
