TP-122 is a bacteriophage cocktail developed by Technophage for the treatment of severe respiratory infections, including ventilator-associated pneumonia (VAP) and hospital-acquired pneumonia (HAP). The therapeutic consists of a cocktail of lytic bacteriophages specifically selected to target three major Gram-negative pathogens: Pseudomonas aeruginosa, Klebsiella pneumoniae, and Acinetobacter baumannii. The Klebsiella-targeting component of the cocktail includes phages from the Drexlerviridae family (formerly classified under Siphoviridae), such as the well-characterized strain vB_KpnS_V6. These phages function by recognizing specific surface receptors on the target bacteria, injecting their viral DNA, and hijacking the host's cellular machinery to replicate. This process culminates in the lysis of the bacterial cell, releasing new virions to infect adjacent bacteria. TP-122 is designed to be administered via inhalation to deliver the phages directly to the site of infection in the lungs, providing a targeted approach against multi-drug resistant (MDR) strains where conventional antibiotics may fail.
