TPF + cetuximab

TPF + cetuximab is a multi-drug therapeutic regimen evaluated by the Grupo Español de Tratamiento de Tumores de Cabeza y Cuello (TTCC) for the treatment of resectable locally advanced laryngeal squamous cell carcinoma. The protocol consists of induction chemotherapy using the TPF triplet—docetaxel (a microtubule stabilizer), cisplatin (a DNA-damaging platinum agent), and fluorouracil (an antimetabolite that inhibits thymidylate synthase)—followed by radiotherapy administered concurrently with cetuximab. Cetuximab is a chimeric monoclonal antibody that targets the extracellular domain of the epidermal growth factor receptor (EGFR), which is frequently overexpressed in head and neck cancers. By blocking ligand binding, cetuximab inhibits downstream signaling pathways that promote tumor cell proliferation, survival, and angiogenesis. This sequential combination is designed to maximize tumor shrinkage and facilitate organ preservation, specifically aiming to improve laryngectomy-free survival in patients who would otherwise require radical surgery.
