TPIV200 + durvalumab is an investigational combination immunotherapy being evaluated for the treatment of platinum-resistant ovarian cancer. TPIV200 is a multi-epitope peptide vaccine targeting the folate receptor alpha (FRα), a protein overexpressed in the majority of ovarian, fallopian tube, and primary peritoneal cancers. The vaccine consists of five T-helper peptides derived from FRα, designed to induce a robust and long-lasting T-cell immune response against tumor cells. Durvalumab (Imfinzi) is a human monoclonal antibody that binds to programmed death-ligand 1 (PD-L1), blocking its interaction with PD-1 and CD80, thereby reversing immune suppression. The combination strategy aims to enhance anti-tumor efficacy by simultaneously priming the immune system against a specific tumor antigen (via TPIV200) and preventing the tumor from evading immune detection (via durvalumab). This specific regimen is being evaluated in a Phase 2 trial sponsored by Memorial Sloan Kettering Cancer Center.
