TPM-01 is a transdermal patch formulation of morphine developed by Avecho Biotechnology (formerly Phosphagenics). It utilizes the proprietary Tocopheryl Phosphate Mixture (TPM) delivery technology, which is a patented combination of mono- and di-tocopheryl phosphates derived from Vitamin E. This technology is designed to enhance the solubility and skin penetration of morphine, allowing for non-invasive, sustained delivery of the opioid through the skin into the bloodstream. The primary goal of TPM-01 is to provide effective analgesia for chronic or post-operative pain while minimizing the gastrointestinal side effects and peak-trough plasma fluctuations typically associated with oral morphine administration. Clinical development reached Phase 2 trials for both chronic and post-operative pain, but the program is no longer active in the developer's current pipeline.
