TQB2102 + benmelstobart + chemotherapy is a combination therapy regimen under development by Chia Tai Tianqing Pharmaceutical Group and Nanjing Shunxin Pharmaceutical Co., Ltd. for the treatment of HER2-positive gastroesophageal adenocarcinoma. TQB2102 is a biparatopic antibody-drug conjugate (ADC) that simultaneously targets two non-overlapping epitopes (ECD2 and ECD4) of the HER2 protein. It is linked to a topoisomerase I inhibitor payload via an enzyme-cleavable linker, designed to enhance HER2 internalization and signaling blockade. Benmelstobart (TQB2450) is a humanized IgG1 monoclonal antibody targeting PD-L1, which blocks the interaction of PD-L1 with PD-1 and CD80 receptors to restore anti-tumor immune responses. The chemotherapy component, typically capecitabine, provides additional cytotoxic activity. This combination is currently being evaluated in Phase II clinical trials to leverage the synergistic effects of HER2-targeted therapy, immune checkpoint inhibition, and traditional chemotherapy in patients with advanced or metastatic disease.
