Overview

TQB2102 + penpulimab + chemotherapy is a combination therapeutic regimen being investigated for the treatment of HER2-positive gastroesophageal adenocarcinoma. The regimen integrates three distinct anti-tumor strategies: TQB2102, a next-generation antibody-drug conjugate (ADC) that bi-specifically targets the ECD2 and ECD4 domains of the Human Epidermal Growth Factor Receptor 2 (HER2) to enhance receptor internalization and deliver a cytotoxic payload; penpulimab (AK105), a humanized IgG1 monoclonal antibody that inhibits the Programmed Cell Death Protein 1 (PD-1) receptor to reactivate the immune system against tumor cells; and conventional cytotoxic chemotherapy. Developed by Chia Tai Tianqing Pharmaceutical Group in collaboration with Akeso (for penpulimab), this combination is currently in Phase II clinical development, aiming to improve outcomes in patients with advanced, recurrent, or metastatic HER2-positive gastric and gastroesophageal junction cancers by leveraging synergistic targeted, immunological, and cytotoxic mechanisms.
