Overview

TQB2916 + penpulimab is a combination of two monoclonal antibodies under investigation for the treatment of solid tumors, including soft tissue sarcoma, undifferentiated pleomorphic sarcoma, and melanoma. TQB2916 is a humanized IgG2 monoclonal antibody that acts as an agonist of CD40, a member of the tumor necrosis factor receptor superfamily involved in immune activation. Penpulimab is a humanized IgG1 anti-programmed cell death protein 1 (PD-1) monoclonal antibody that blocks PD-1-mediated immune checkpoint signaling to enhance anti-tumor immunity. Penpulimab has been approved in China for relapsed or refractory classic Hodgkin’s lymphoma and first-line treatment of nasopharyngeal cancer and squamous non-small cell lung cancer; it is marketed under the brand name ANNIKO. The combination aims to synergistically activate immune responses against tumors by both stimulating antigen-presenting cells via CD40 and releasing PD-1-mediated inhibition on cytotoxic lymphocytes[1][2][5][7].
