**TR-12@LGMN** is a preclinical dissolving microneedle therapeutic cancer vaccine designed for melanoma immunotherapy. The quadrilateral polyvinylpyrrolidone-polyvinyl alcohol microneedle matrix co-delivers granulocyte-macrophage colony-stimulating factor and liposomes containing tyrosinase-related protein 2 peptide, the Toll-like receptor 7/8 agonist resiquimod, and interleukin-12 mRNA. Following intradermal dissolution, granulocyte-macrophage colony-stimulating factor recruits dendritic cells, while the liposomal payload promotes antigen cross-presentation, dendritic-cell maturation, and interleukin-12-mediated T-cell activation. In B16F10 melanoma prevention and treatment models, it induced antigen-specific cytotoxic and memory T-cell responses and durable antitumor immunity.
