tramadol + acetaminophen + naloxone

This investigational combination therapy consists of oral tramadol/acetaminophen (marketed as Tramacet) and an intravenous infusion of ultra low-dose naloxone. It is being developed by researchers at the London Health Sciences Centre Research Institute and the Lawson Research Institute of St. Joseph's Health Care London. The regimen is designed as an opioid-sparing approach for managing postoperative pain in elderly patients (70 years or older) undergoing total knee or hip arthroplasty. Tramadol acts as a weak mu-opioid receptor agonist and serotonin/norepinephrine reuptake inhibitor, while acetaminophen provides non-opioid analgesia. The addition of ultra low-dose naloxone, a mu-opioid receptor antagonist, is intended to reduce opioid-induced side effects such as nausea, vomiting, and sedation without compromising the analgesic efficacy of the combination.
