Overview

Tramadol + dexamethasone is a combination therapy being investigated for the management of postoperative pain, specifically following knee arthroscopy. This formulation combines tramadol, a synthetic opioid analgesic with a dual mechanism of action, and dexamethasone, a potent synthetic glucocorticoid. Tramadol provides analgesia by acting as a mu-opioid receptor agonist and by inhibiting the reuptake of serotonin and norepinephrine, thereby modulating descending pain pathways. Dexamethasone acts as an anti-inflammatory agent by binding to glucocorticoid receptors, which can reduce local inflammation and potentially prolong the analgesic effect of local or regional treatments. In this specific clinical application, the combination is administered intraarticularly at the end of surgery to provide localized, effective pain relief and reduce the overall consumption of systemic analgesics.
