Overview

The regimen consists of a standard dose of the MEK inhibitor trametinib (2 mg once daily) and a low dose of the BRAF inhibitor dabrafenib (50 mg twice daily). In this specific clinical context, trametinib serves as the primary therapeutic agent by inhibiting MEK1 and MEK2 kinases within the MAPK signaling pathway to suppress tumor growth. The low-dose dabrafenib is added not for its anti-tumor activity—as the patients have BRAF wild-type tumors—but specifically to mitigate trametinib-related skin toxicities, such as acneiform rash, by modulating paradoxical MAPK pathway activation in keratinocytes.
