Overview

A combination therapy consisting of trametinib, a MEK inhibitor, and digoxin, a cardiac glycoside. This combination has been investigated for the treatment of BRAF wild-type metastatic melanoma, particularly in patients refractory or intolerant to immune checkpoint blockade. Preclinical studies showed that cardenolides (like digoxin) synergize with MEK inhibitors to promote regression of patient-derived xenografts regardless of BRAF mutation status. The combination appears particularly effective in NRAS mutant melanomas.
