Overview

Trametinib + paclitaxel is a combination therapy investigated by Memorial Sloan Kettering Cancer Center for the treatment of anaplastic thyroid cancer (ATC). Trametinib is an orally bioavailable, small molecule, highly selective allosteric inhibitor of mitogen-activated protein kinase kinase 1 (MEK1) and MEK2, which are key components of the MAPK/ERK signaling pathway often dysregulated in thyroid cancers. Paclitaxel is a taxane-based chemotherapy agent that stabilizes microtubules, preventing their disassembly and thereby inhibiting cell division (mitosis). This specific combination was evaluated in a Phase 2 pilot study (NCT03085056) to assess its safety and efficacy in patients with metastatic or locoregional ATC, aiming to provide a therapeutic option for a disease with a very poor prognosis. While trametinib is FDA-approved in combination with dabrafenib for BRAF V600E-mutant ATC, this regimen explores its utility with cytotoxic chemotherapy.
